Hi Daniel,
if you dont have a mapfile, create one (see 
http://mapserver.gis.umn.edu/docs/reference/mapfile)
then simply follow the instructions of 
http://www.cartoweb.org/cwiki/HowToCreateANewProject, especially "Your Own 
Data" chapter
