It sounds to me as if Mapserver can not locate and read your data "region". 
Double check which system path is in you map file and where your actual data
(shapefile) is located. Also check whether spelling and capital/lower case
letters of the data tag in the map file and your shape file name match.

when I customize the project to add my own shape file instead of it and run
the project
