Mark,
I don't know what you're backend is but I can report that Oracle Spatial works very well. We use it extensively.
ESRI has announced its intention to discontinue SDE in favor of its new SDS product (Spatial Data Server). I don't know that I would be planning on SDE long term

Subject: [MS4W-Users] SDE workgroup vs shapefile performance thoughts
Hello,
We are considering using SDE workgroup as a data source for Mapserver. Does anyone have any comments about the performance or usability of SDE Workgroup compared to shapefiles?
Thanks
