yes, but dont forget the path must be either relative to the map's SHAPEPATH parameter, or absolute.
maybe you were just missing the : ?
DATA "D:/temp/test"
the error returned by mapserver is usualy very clear

is it possible to set in map file a layer outside the folder "DATA"?
Ex:
....
DATA "test" the layer test.shp is inside Data folder
....
...
DATA "D/temp/test" the layer test.shp is outside Data folder, inside temp.
....
Directory error?
